Active Ingredients:

• Losartan 20 mg (Angiotensin II receptor blocker, ARB)

• Hydrochlorothiazide 5 mg (Thiazide diuretic)

• Amlodipine 12.5 mg (Calcium channel blocker)

Therapeutic Class:
Triple combination antihypertensive – ARB + Thiazide diuretic + Calcium channel blocker

Indications:

• Hypertension (high blood pressure) in adults, particularly when monotherapy or dual therapy is insufficient.

• Used to achieve better blood pressure control and reduce cardiovascular risk.

Dosage:

• Usual adult dose: One tablet once daily, preferably in the morning.

• Can be taken with or without food.

• Dose adjustments may be required based on blood pressure response, kidney function, and tolerance.

Mechanism of Action:

• Losartan: Blocks angiotensin II type 1 (AT1) receptors, preventing vasoconstriction and aldosterone secretion.

• Hydrochlorothiazide: Increases sodium and water excretion, reducing blood volume and lowering blood pressure.

• Amlodipine: Blocks calcium channels in vascular smooth muscle, causing vasodilation and reducing peripheral resistance.

• The combination provides synergistic antihypertensive effects through complementary mechanisms.

Precautions:

• Contraindicated in:

  • Pregnancy and breastfeeding

  • Severe renal impairment

  • Hypersensitivity to any component

• Use with caution in:

  • Electrolyte imbalances (especially potassium)

  • Liver or kidney impairment

  • Heart failure or volume depletion

• Monitor blood pressure, kidney function, and electrolytes regularly.

Possible Side Effects:

• Dizziness or lightheadedness

• Fatigue

• Peripheral edema (swelling of hands/feet)

• Electrolyte disturbances (low sodium, high potassium)

• Flushing or headache

• Rare: kidney impairment, angioedema, hypotension
