Medication Name: Dapoxetine

Pharmacological Class:

• Selective Serotonin Reuptake Inhibitor (SSRI) – short-acting

Indications:

• Premature Ejaculation (PE) in adult men aged 18–64 years

Mechanism of Action:
Dapoxetine inhibits the reuptake of serotonin in the presynaptic neuron, increasing serotonin levels in the synaptic cleft. This enhances neurotransmission in pathways that delay ejaculation, improving control and extending the time before ejaculation during sexual activity.

Dosage:

• Initial dose: 30 mg taken 1 to 3 hours before sexual activity.

• If the effect is insufficient and side effects are tolerable, the dose may be increased to 60 mg.

• Maximum frequency: Once in 24 hours.

Precautions:

• Not intended for continuous daily use — only “on-demand.”

• Avoid in patients with heart disease, severe liver impairment, or history of syncope or seizures.

• Avoid concurrent use with MAO inhibitors, SSRIs, tricyclic antidepressants, thioridazine, or CYP3A4 inhibitors (e.g., ketoconazole, erythromycin).

• Avoid alcohol — increases the risk of fainting or dizziness.

Common Side Effects:

• Nausea

• Dizziness

• Headache

• Insomnia

• Fatigue

• Diarrhea

• Anxiety or nervousness

Onset of Action:

• Works within 1–3 hours after ingestion.

Duration of Action:

• Effect lasts approximately 6–8 hours.

Note:
Dapoxetine is the only SSRI specifically approved for premature ejaculation. It should not be used for depression or anxiety disorders. Always start with the lowest effective dose and assess individual response and tolerance.
