Dosage Forms:

Indications:
Treatment of erectile dysfunction (ED) in adult men.
Mechanism of Action:
Vardenafil selectively inhibits the PDE5 enzyme, which degrades cyclic guanosine monophosphate (cGMP) in the corpus cavernosum.
Sexual stimulation triggers nitric oxide release, which increases cGMP and causes smooth muscle relaxation and enhanced penile blood flow, leading to erection.
Vardenafil enhances this effect but does not induce erection without sexual stimulation.
Dosage and Administration:

Recommended starting dose: 10 mg, taken 25–60 minutes before sexual activity.

May be increased to 20 mg or decreased to 5 mg depending on efficacy and tolerability.

Maximum: One dose per 24 hours.

Can be taken with or without food, but high-fat meals may delay onset.
Contraindications:

Concurrent use with nitrates or nitric oxide donors (risk of severe hypotension).

Severe hepatic impairment.

Known hypersensitivity to vardenafil or any excipients.
Precautions:

Use cautiously in patients with cardiovascular disorders (e.g., unstable angina, recent stroke or myocardial infarction).

Avoid in men with retinitis pigmentosa or uncontrolled hypertension/hypotension.

Avoid use with strong CYP3A4 inhibitors (e.g., ketoconazole, ritonavir).

Alcohol may reduce the ability to achieve an erection.
